摘要
目的观察油酸致伤后大鼠肺组织细胞外信号调节激酶(ERK)和磷酸化ERK(P-ERK)与肿瘤坏死因子(TNF-α)及白介素-6(IL-6)释放的关系。方法健康雄性SD大鼠60只,随机分为油酸1h组,油酸2h组,油酸4h组,油酸24h组和对照组,观察各组肺组织病理学改变,免疫组化法和蛋白定量法(Western blot)测定肺内ERK,P-ERK表达,放免法检测血清、BALF、肺组织匀浆液中TNF-α及IL-6水平。结果油酸组大鼠支气管、肺泡上皮细胞、肺血管内皮细胞、肺泡巨噬细胞上表达ERK和P-ERK明显增多,血清、BALF、肺组织匀浆液中TNF-α及IL-6水平明显升高,TNF-α与ERK和P-ERK表达峰值时间均为1h,IL-6表达峰值时间为4h。结论ERK信号通路参与调节油酸型急性肺损伤大鼠IL-6的释放过程,与TNF-α为互相促进的关系。
Objective To observe the ralationship of extracellular signal regulated protein kinase (ERK) and phosphorylating ERK with TNF - αtand IL - 6 in rats with acute lung injury induced by oleic acid. Methods 60 healthy male SD rats randomly divided into 5 groups: Oleic acid 1 h group, Oleic acid 2 h group, Oleic acid 4 h group, Oleic acid 24 h group and control group. Pathology changement of the lung was investigated. ERK and P - ERK expression condition was determined by Western blot and immunohistochemical method. Levels of TNF -α and IL -6 in the rat's serum, the lung tissue and BALF was examined by the Radioimmunnol method. Results ERK and P - ERK in bronchus epithelial cells, alveolar epithelial cells, pulmonary vessel endothelial cells and macrophage cell was highly expressed in the lung of OG rats ,The level of TNF -α and IL- 6 in serum, the lung tissue and BALF increased obviously after rat being injected oleic and the peek value time of ERK, P- ERK and TNF -α was same in 1 h after injured, the peek value time of IL - 6 was 4 h. Conclusions ERK signal transaction path participated IL - 6 releasion process while with TNF -α for mutual promotive relationship.
