转化生长因子-β_1体外诱导气道上皮细胞向肌纤维母细胞转分化 被引量:4

摘要 目的探讨TGF-β1体外诱导气道上皮细胞表型向肌纤维母细胞转分化的可能性。方法TGF-β1(10μg/L)处理气道上皮细胞株16HBE-14o,显微镜下实时观察形态学变化,免疫染色方法检测E-cadherin,α-SMA和F-actin的表达,RT-PCR方法检测E-cadherin,α-SMA mRNA的表达。结果TGF-β110μg/L作用3 d,气道上皮细胞株16HBE-14o失去正常的立方形,变得肥大,胞体拉长;上皮细胞特征性标志E-cadherin表达减弱,胞浆中出现肌纤维母细胞表型标志物α-SMA,F-actin聚合形成张力纤维沿着细胞长轴分布。结论本研究提示,气道上皮细胞在TGF-β1的作用下经历了表型改变向肌纤维母细胞转分化,可能作为肺肌纤维母细胞的一个新的来源,从而参与哮喘气道重塑、气道高反应性的发生发展。
