河虾中主要过敏原组分鉴定及分离纯化 被引量:8

摘要 目的鉴定并分离纯化河虾中引起过敏反应的主要蛋白组分。方法河虾蛋白浸液皮下注射建立小鼠过敏模型,获得高效价特异性IgE血清用于免疫印迹分析。用饱和硫酸铵分段盐析和DEAE离子交换层析纯化目的蛋白。dot-ELISA及竞争ELISA检测纯化目的蛋白的过敏原活性。结果河虾蛋白浸液皮下注射成功建立了虾过敏小鼠模型。免疫印迹分析显示虾中有一分子量36 kD的蛋白组分为主要过敏原。用硫酸铵分级沉淀和DEAE离子交换层析的方法成功分离到目的蛋白。dot-ELISA证明目的蛋白能与特异性IgE反应。竞争ELISA检测纯化目的蛋白与河虾蛋白浸液竞争结合特异性IgE的活性,最大竞争抑制率可达60%。结论本实验成功鉴定并分离到河虾中一分子量为36 kD的主要过敏原组分,为过敏原的分析研究提供了一套比较完整的方法。
