大白菜胞质雄性不育系育性相关线粒体DNA片断的克隆及*序列分析 被引量:7

Cloning and Sequencing of a Mitochondrial DNA Fragment Related to Cytoplasmic Male Sterile Line of Chinese Cabbage

摘要 利用PCR技术从大白菜(Brassica campestrisL.ssp.Penkinsis)新型胞质雄性不育材料CMS7311的线粒体基因组中扩增出与细胞质雄性不育相关的片断,并克隆测序。序列分析表明,该片断和报道的Poli-ma-orf224序列完全相同,没有发生变异,说明CMS7311的不育机理与PolimaCMS相似,也反映了细胞质雄性不育变异的保守性。 Based on the nucleotide sequence of "Polima" CMS associated specific fragment orf224 in Brassica napus, two primers were synthesized and applied to polymerase chain reaction with the templates of mitochondrial DNA of new CMS line CMS7311 of Chinese cabbage (Brassica campestris L. ssp. Penkinsis) and it's maintainer line 7311. A special amplified fragment obtained only in CMS line CMS7311 by PCR was cloned into pMD18 T vector. Sequence analysis revealed that this fragment has no variation compared with Polima-orf224. This means that there has similar sterile mechanism in CMS7311 with Polima CMS and the variation of CMS in Brssica napus is conservative.
