摘要
以狗头枣和骏枣试管苗为材料,研究了TDZ和6-BA在枣树试管苗继代培养中的效应并筛选了适宜组合。结果表明,在枣试管苗继代培养中,TDZ与6-BA分别与IBA配合使用时,附加6-BA 1.2~1.6mg/L比较适合;TDZ的活性高于6-BA。其诱导的不定芽比6-BA的多但芽曲质量显著降低。二者结合使用则可显著促进不定芽的增殖并提高成苗质量,在CY+TDZ 0.005~0.01mg/L+6-BA1.0mg/L+IBA0.2mg/L的组合培养基上可获得最佳繁殖效果。TDZ与IBA配合使用时能抑制试管枣苗生根,但可促使茎段愈伤组织的诱导。说明TDZ具有强烈地刺激细胞分裂的作用,并且在枣试管苗继代培养中与6-BA具有很好的协同促进作用。
Use test-tube seedlings of Chinese date Goutouzao and Junzao as explants, effect of TDZ and 6-BA on subculture of Chinese date were studied and selected its suitable composition in subculture. The results showed that in subculture added 1. 241.6 mg/L 6- BA was suitable, then them respectively together with IBA. Activity of TDZ was higher, and it induced bud, more bud, but quality of seedlings cut down, as compare with 6- BA. TDZ and 6-BA blended with IBA can remarkable pro mote propagate of buds and quality of grow into seedlings; The best effect obtained on the medium containing CY + 0. 005-0.01 mg/L TDZ + 1.0 mg/L 6- BA + 0.2 mg/L IBA. Then TDZ blended with IBA can check growth of root of test-tube seedlings but can promote induction of ealli in stem segment. It showed that the TDZ have effects of strongly stimulated cell division and with 6-BA have best cooperation effects.
