摘要
目的:观察EPA和DHA对人胃癌BGC-823细胞凋亡信号传导的影响。方法:采用MTT法测定肿瘤细胞抑制率,流式细胞检测肿瘤细胞凋亡率和bcl-2、Gα、PKCβⅡ基因蛋白的表达。结果:45μg/ml和48h时间点的EPA或DHA可显著抑制肿瘤细胞的生长(P<0.001),凋亡率明显升高(P<0.001),bcl-2基因蛋白表达下降(P<0.05),Gα和PKCβⅡ基因蛋白表达明显下降(P<0.001)。结论:ω-3脂肪酸可诱导人胃癌BGC-823细胞发生凋亡,信号传导途径Gα-PKCβⅡ的改变在细胞凋亡中起到重要的作用。
Objective: To assess the effects of EPA and DHA on signal transduction for apoptosis of the human gastric BGC-823 cells in vitro. Methods: Inhibitive rate of the cells was determined by MTT assay. Apoptotic rate and expression of bcl-2, Gα and PKCβⅡ proteins were determined using flow cytometry. Results: After treatment with 45μg/ml of EPA or DHA and at the exposed point of 48h, the growth of cells was markedly inhibited (P〈0.001), and apoptotic rate was markedly increased (P〈 0.001). Expression of bcl-2 was decreased (P〈0.05), and expression of Gα and PKCβⅡ protein were markedly decreased (P〈0.001). Conclusions: The ω-3 fatty acids can induce apoptosis of human gastric BGC-823 cells. Gα and PKCβⅡ signaling may play an important role in apoptosis of the cells.
