摘要
目的探讨胰岛素样生长因子系统(IGFs)在体外培养的胎盘绒毛滋养层细胞中的基因表达及其作用。方法采用逆转录-聚合酶链反应(RT-PCR)技术检测58例体外培养的胎盘绒毛滋养层细胞中IGF-ImRNA、IGF-IImRNA、IGF-IRmRNA、IGF-IIRmRNA、IGFBP-1mRNA、IGFBP-2mRNA、IGFBP-3mRNA、IGFBP-4mRNA、IGFBP-5mRNA、IGFBP-6mRNA等10种胰岛素样生长因子的基因表达。结果58例体外培养的胎盘绒毛滋养层细胞均有IGF-ImRNA、IGF-IImRNA的基因表达;56例存在IGF-IRmRNA、IGF-IImRNA、IGFBP-3mRNA的基因表达;2例同时检测出上述10种基因。结论IGFs可能在早期细胞滋养层的侵入、增殖分化、胎盘形成和胚胎生长发育过程中以自分泌或旁分泌方式发挥重要的调节作用。
Objective To investigate the gene expression and action of insulin - like growth factors (IGFs) in human villous trophoblast cells of early pregnancy in vitro. Methods Reverse transcfiptase polymerase chain reaction ( RT - PCR) was used for determining the expression of GFs(IGF - ⅠmRNA, IGF - ⅡmRNA, IGF - ⅠRmRNA, IGF - ⅡRmRNA, IGFBP - 1mRNA, IGFBP - 2mRNA, IGFBP- 3mRNA, IGFBP-4mRNA, IGFBP- 5mRNA, IGFBP- 6mRNA) messenger RNA in human villous trophoblast cells of 58 early pregnancy cases in vitro. Results The expression of IGF - ⅠmRNA, IGF - ⅡmRNA was detectable in human villous trophoblast cells of 58 early pregnancy cases in vitro, IGF-ⅠRmRNA, IGF-ⅡmRNA and IGFBP- 3mRNA were detectable in 56 early pregnancy cases and ten IGFs were detectable in 2 early pregnancy cases. Conclusion IGFs appear to play important regulation roles in early invasion, proliferation and differentiation of cytotrophoblast, and in the formation of placenta and the development of embryo via autocfine or paracfine way.
