摘要
目的研究肿瘤坏死因子α(TNF-α)和肿瘤坏死因子受体Ⅰ(TNFR-Ⅰ)在胆管癌中的表达及与胆管癌分化程度、浸润深度、淋巴结转移等之间的关系。方法收集胆管癌手术标本48例,常规HE染色分组后,用SP免疫组织化学法检测TNF-α和TNFR-Ⅰ的表达情况。结果TNF-α主要在胆管癌细胞的胞浆中表达,阳性率为25%(12/48),其表达强度与胆管癌分化程度、浸润深度及淋巴结转移无关,P>0.05;TNFR-Ⅰ主要在胞膜和胞浆中表达,阳性率为75%(36/48),其表达强度与胆管癌分化程度、淋巴结转移无关,P>0.05,但与浸润深度呈负相关,P<0.01。TNF-α与TNFR-Ⅰ二者表达强度比较差异有统计学意义(P<0.01)。结论TNF-α和TNFR-Ⅰ在胆管癌细胞中均有表达,而且TNF-α的表达明显低于TNFR-Ⅰ。TNF-α的表达水平与胆管癌的分化、浸润、转移等生物学行为无关。TNFR-Ⅰ的表达水平与胆管癌的分化程度、有无淋巴结转移等无关,但与浸润深度呈负相关。
Objective To investigate the expression of tumor necrosis factor- α(TNF-α)and necrosis factor receptor -Ⅰ (TNFR -Ⅰ) in cholangiocarcinoma (CCA) tissues, and the relations between the expression and the classify, invasion, as well as the differentiation of CCA. Method TNF-α and TNFR-Ⅰ expression in tissues were immunohistochemically analyzed from resected (CCA, which had been divided into groups by hematoxylin and eosin (HE) stain. Result Both TNF-α and TNFR-Ⅰ could expressed in CCA. TNF-α mainly expressed in cytoplasm of CCA cell, and the positive rate was 25% (12/48). The expression of TNF-α was not correlated with the tumor differentiation, depth of invasion and lymph node metastasis (P〉0.05). TNFR-Ⅰ expressed in both cytoplasm and cytomembrane of CCA cell, the positive rate was 75% (36/48). There was no significant difference between the TNFR-Ⅰ expression and tumour dedifferentiation, as well as metastasis (P 〉 0.05), the expression of TNFR-Ⅰ was significantly difference in depth of invasion (P〈 0.01). There was significant difference between TNF-α expression and TNFR-Ⅰexpression(P〈0.01). Conclusion tissues can express TNF-α and TNFR-Ⅰ. TNF-α expressed obviously lower than that of TNFR -Ⅰ does in CCA. The expression of TNF-α in CCA has not correhted with the tumors differentiated, depth of invasion and lymph node metastasis. The expression of TNFR -Ⅰ has no significant difference between tumor differentiation and metastasis, but it has significantly difference in depth of invasion.
