青春双歧杆菌细胞壁对T_(24)细胞的抑制作用研究 被引量:1

摘要 目的探讨青春双歧杆菌细胞壁对小鼠腹腔巨噬细胞功能的影响对人移行细胞膀胱癌细胞的体内外抑制作用。方法采用青春双歧杆菌细胞壁小鼠腹腔注射,以ELISA法测定腹腔巨噬细胞产生的细胞因子IL-2的含量,建立膀胱癌小鼠模型,腹腔注射双歧杆菌细胞壁,剥离瘤体,计算抑瘤率;采用MTT法测定双歧杆菌细胞壁体外对人移行细胞膀胱癌细胞的抑制作用。结果青春双歧杆菌细胞壁注射组腹腔巨噬细胞产生的IL-2的含量明显高于对照组,两者比较具有非常显著性差异(P<0.01);双歧杆菌细胞壁注射组可以使小鼠局部肿瘤体积减小,肿瘤重量减轻与对照组之间比较有非常显著性差异(P<0.01)。不同浓度双歧杆菌细胞壁在体外均能抑制T24细胞生长,且浓度越大,对该细胞的抑制作用越强。结论青春双歧杆菌细胞壁成分能激活小鼠腹腔巨噬细胞,使之分泌多量的IL-2,并且在体外能抑制人移行细胞膀胱癌细胞的生长。 Objective To explore the immunologic regulation of the bifidobacterium adolescence cell wall(BCW) on function of macrophage in mice and tumor inhibition of BCW on human transitional cell carcinoma of bladder(BTCC) inside and outside of body. Methods BCW were injected into abdomen of KM mice,the level of IL--2 which produced by macrophage was determined by ELISA reagent box. The role of tumor inhibition was determined by MTT method^BCW was injected in abdomen after establishing human transitional cell carcinoma of bladder, peel off the tumor tissue, weight, calculated the tumor-inhibition rate. Results The level of IL-2 of BCW injection group was higher than of control group (P〈 0. 01 ) ; The tumor-inhibition became stronger with the increase of BCW concentration; There was significant different between BCW injected group and control group (P〈0.01). Conclusions ECW can activate macroohage to secrete more IL-2 ; BCW can inhibit the growth of BTCC inside and outside body
