表皮生长因子抑制小脑颗粒细胞凋亡的作用 被引量:1

摘要 目的了解表皮生长因子(EGF)对小脑颗粒细胞(rCGC)的保护作用。方法在细胞培养24小时的rCGC中,分别加入EGF及一氧化氮酶抑制剂NG-硝基-L-精氨酸(NG-nitro-L-arginine methylester,NAME)。再经24小时二氧化碳恒温培养后,通过光学显微镜观察它们对rCGC产生的影响。结果EGF能显著减少培养细胞的损失,NAME可抑制EGF的作用。结论EGF通过一氧化氮酶抑制细胞凋亡而对小脑颗粒细胞产生保护作用。 Objective To analyze the effect(s) of nimc oxide syntbase(NOS) which may relate to the epithelium growth factor(EGF) mediated neurotrophism on rat cerebellar granule cell(rCGC) in culture. Methods rCGC culture had been harvested and then incu bated for 24 hours before testing different reagents, such as EGF and NAME, a biDS inhibitor. The rCG C cultures were kept in incubator in sealed containers for another 24 hours before cells were collected and counted by using hemocytometer. Results EGF bad been found neurotrophic effect to culture since it significantly reduces cell death in culture. NAME could eliminate the EGF mediated neurotrophic effect to the rCGC. Conclusion NOS is necessary in the mechanism of EGF - mediated neurotrophism to rCGC culture.
