摘要
目的探讨芹菜素在生殖轴调控中的作用及作用部位。方法通过阴道涂片选用情期成年雌性大鼠50只,随机分为五组,分别侧脑室注入人工脑脊液、二甲基亚砜、芹菜素10-5M、hCG及芹菜素10-5M+hCG,各组再分为3d组、5d组。分别于侧脑室注射后的3d、5d断头取血、卵巢及垂体,应用放射免疫方法分别测定血清、卵巢组织及垂体组织中FSH、LH、E2及P含量,以观察浓度为10-5M的芹菜素对生殖轴的作用。结果1.人工脑脊液组与二甲基亚砜组无明显差异。2.单用10-5M芹菜素组与人工脑脊液组相比FSH、LH、E2、P的含量数值小,其差异有显著性。3.单用hCG组与人工脑脊液组相比FSH、LH的含量数值小,而E2、P的含量数值大,其差异均有显著性。4.10-5M芹菜素与hCG合用组与单用10-5M芹菜素组相比FSH及LH的含量数值小,而E2、P的含量数值大,其差异均有显著性。5.10-5M芹菜素与hCG合用组与单用hCG组相比FSH、LH的含量数值大,而E2、P的含量数值小,其差异均有显著性。结论芹菜素在下丘脑水平发挥作用,进而作用于腺垂体、卵巢。10-5M芹菜素能够抑制FSH、LH的合成,从而抑制E2、P的分泌。10-5M芹菜素与hCG合用时可表现出抗雌激素的作用。
Objective The main idea of this study was to investigate the effect and the position of apigenin on the reproductive axis of female rat in vitro. Methods Adult female S-D rats were injected ACSF, DMSO, 10^-5mol/L Api, 10^-5mol/L Api + hCG, through lateral brain ventricle and decapitated to take blood, pituitary and ovary.FSH, LH, E2 and substance P in serum, pituitary and ovary were determined by radiommunassay. Results (1) The ACSF group wasn' t significant deviation than the DMSO group. (2) FSH, LH, E2 and P of the Api group were significance level than the ACSF group. (3) FSH, LH of the DMSO group were significant decrease than the ACSF group, but E2, P were significant figures. (4) FSH, LH of the Api + hCG were significant decrease than.the Api group, but E2, P were significant figures. (5) FSH, LH of the Api + hCG were significant figures than the hCG group, but E2, P were significant level. Conclusions Apigenin inhibits FSH, LH, E2 and substance P release in the level of hypothalamus. 10^-5mol/L Api + hCG have beneficial for morphology of hormone.
