摘要
目的探讨枸杞多糖(LBP-X)诱导人类慢性髓系白血病K562细胞株凋亡。方法用吖啶橙(AO)荧光染色观察细胞结构的变化,用MTT比色法测定抑制率,琼脂糖凝胶电泳和流式细胞仪检测法分析细胞凋亡。结果LBP-X可明显抑制K562细胞的生长,凋亡率呈时间和剂量依赖性。LBP-X作用48h后,荧光显微镜下可见细胞核不规则,浓缩成大小不等的团快,核碎裂呈新月形改变,琼脂糖凝胶电泳可见DNA梯带。流式细胞仪分析图上可见明显的凋亡。结论LBP-X能诱导K562细胞的凋亡,并呈一定的时间浓度依赖关系。
Objective To study the effect of lycium bararum polysaccharide(LBP - X) on apoptosis of human chronic myeloid leukemia K562 cells. Methods Morphological changes were observed by AO fluorescent staining. Inhibition rate of cell proliferation were stuidied by MTT assay. Apoptosis was analyzed by agarose gel electrophoresis and flow cytometry. Results The growth of K562 cells were inhibited remarkably. Apoptosis of K562 cells depended on dose and time. LBP induced the nuckei of K562 with anomalistic, condensed and fragmentized for 8 hours. Agarose gel electrophoresis of DNA revealed DNA ladder , K562 cells extposed to LBP- X showed apoptotic peaks. Conclosions LBP- X can induce apoptosis of K562 cells, and the inhibition of cell character depended on dora and time.
