γ射线和微波复合照射小鼠骨髓细胞凋亡形态学特点及Bax、Bcl-2蛋白表达定量分析 被引量:9

摘要 目的研究60Coγ射线(以下简称γ线)与微波复合照射后小鼠骨髓细胞的凋亡情况及凋亡相关基因bax、bcl-2表达规律,以探讨复合照射对造血组织的损伤机制。方法用光镜及透射电镜观察复合照射后不同时间点小鼠骨髓细胞凋亡的变化,用原位末端标记法检测复合照射后6 h骨髓细胞凋亡率,用免疫组化和图象分析技术检测骨髓细胞Bax、Bcl-2蛋白表达。结果γ线与微波复合照射后出现典型的凋亡改变,以照后6 h最为明显;微波组骨髓细胞凋亡率仅轻微升高,而γ线组和复合组则显著升高(p<0.01),同单纯γ线组相比,复合组凋亡率升高更明显(p<0.05)。照后初期Bax表达增强、Bcl-2表达降低,与γ线组相比,复合组照射后6 h Bax表达增强更为明显(p<0.05)。结论γ线与微波复合照射后小鼠骨髓细胞发生凋亡,微波可进一步促进γ线对骨髓细胞凋亡的诱导作用。Bax和Bcl-2协调发挥作用是复合照射导致骨髓细胞发生凋亡且较单伤加重的机制之一。 Objective To explore apoptosis of bone marrow hematopoietic cells of mouse irradiated by γrays combined with microwave, and investigate the expression of Bax and Bcl-2. Methods Apoptosis of bone marrow hematopoietic cells in mice irradiated with γrays irradiation combined with microwave was detected by in situ end labeling, electron microscopy (EM) and light microscopy. The expressions of Bax, Bcl-2 proteins were detected by inmunohistochemical and image analysis methods. Results γray combined with microwave could induce apoptosis of bone marrow cells, especially in 6 h after radiation. The amount of apoptosis cells in both γray irradiation group and combined radiation group were more than that of control group. Moreover, the amount of apoptosis cells in combined radiation group was more than that in γray irradiation group. In the early days after irradiation, Bax protein expression was raised dramatically as while as the expression of Bcl-2 was decreased. Conclusion γray combined with micro-wave could induce apoptosis of bone marrow hematopoietic cells, and microwave could enhance the effect Of γray. The expressions of Bax and Bcl-2 in harmony may play a role in apoptosis of bone marrow cells.
