摘要
目的探讨水通道蛋白4(AQP4)及基质金属蛋白酶2,基质金属蛋白酶9(MMP-2, MMP-9)在高血压性脑出血后的早期表达,为临床上防止脑组织进一步损伤提供实验依据。方法应用半定量RT-PCR方法检测高血压性脑出血患者出血后24 h内血肿周围脑组织中AQP4及MMP-2、 MMP-9的表达。结果 AOP4、MMP-2及MMP-9在高血压脑出血血肿周围组织中其相对表达量明显升高,相对表达量分别为0.517±0.028、0.502±0.054和1.003±0.013,与正常对照组0.021±0.004、 0.271±0.103和0.239±0.021相比差异均具有显著性(P<0.001)。结论 AQP4、MMP-2及MMP-9的早期上调表达在高血压性脑出血后早期脑水肿的发生、发展过程中具有重要的作用。
Objective To explore the early expressions and their roles of aquaporin-4 (AQP4), matrix metalloproteinases 2 and 9 (MMP-2 and MMP-9, respectively) in the development of brain edema after hypertensive intracerebral hemorrhage for providing the experimental evidence for prevention of brain injury. Methods Hemi-quantitative RT-PCR was used to detect the mRNA expressions of AQP4, MMP-2 and MMP-9 in 24 h after hypertensive intracerebral hemorrhage. Results The relative expressions of AQP4, MMP-2 and MMP-9 mRNA increased significantly in 24 h after hypertensive intracerebral hemorrhage as compared with the controls (0.517±0.028 vs 0.021 ±0.004, 0.502±0.054 vs 0.271 ±0.103, and 1.003±0.013 vs 0.239±0.021, respectively, P〈0.001). Conclusion The early up-regulated expressions of MMP-2, MMP-9 and AQP4 might play an important role in the occurrence and development of brain edema after hypertensive intracerebral hemorrhage.
