糖尿病大鼠肾病变过程中核因子-κB及诱导型一氧化氮合酶的表达 被引量:6

摘要 目的:探讨糖尿病肾病变与核因子-κB活化及诱导型一氧化氮合酶(iNOS)表达的关系。方法:雄性SD 大鼠60只,随机分成对照1、3、6个月组和糖尿病1、3、6个月组,每组10只。分别于1、3和6个月末处死大鼠,留取肾标本,观测肾脏病理改变并用免疫组化法分析NF-κB及iNOS表达。结果:肾病理结果显示,与对照组相比, 糖尿病1、3、6个月组肾病理改变逐步加重;免疫组化分析结果显示NF-κB和iNOS表达在糖尿病各组均明显增强 (P<0.05)。结论:NF-κB及iNOS伴随着肾病理学改变而持续活化,可能在糖尿病肾病(DN)发生中有重要作用, 可作为反映进行性肾损害的参考指标。 Objective: Activation of the transcription factor nuclear faetor-kappaB ( NF-κB ) has been suggested to participate in chronic disorders, such as diabetes and its complications. In contrast to the short activation of NF-κB , we observed a long-lasting activation of NF-κB and iNOS in the kidneys of diabetic rats. Methods: Sprague-Dawley rats were randomly divided into control group and diabetic group. The pathological changes in the kidneys of diabetic rats were observed at 1, 3 and 6 months respectively. The activity of NF-κB and iNOS was detected by immunohistoehemistry. Results: The change of histopathology was severer at 6 months than at 3 months and 1 month in the kidneys of diabetic rats. A comparable increase in NF-κB p65 antigen was observed in the kidney cells especialy in vascular endothelial, cells of diabetic rats, and the expression of iNOS in renal tissues of diabetic rats was higher than that of normal control. Conclusion : NF-κB and iNOS may play an important role in the pathogeneis of diabetic rats, and the expression of them may be considered as a predictor of progressive renal injury in diabetic rats.
