Toll样受体3参与双链RNA对小鼠妊娠结局的影响 被引量:1

摘要 目的:探讨Toll样受体3(TLR3)与双链RNA(dsRNA)相互作用对BALB/c和NOD/SCID小鼠妊娠结局的影响。方法:比较经过和未经预先TLR3阻断条件下同基因妊娠的BALB/c和NOD/SCID小鼠在dsRNA刺激后的胚胎吸收率(RR),并采用流式细胞术检测CD45+TLR3+细胞和CD45+CD80+细胞在CD45+细胞群中、以及CD8α+CD80+细胞在CD8α+细胞群中的百分率。结果:在dsRNA刺激下BALB/c小鼠RR显著升高,而对NOD/SCID小鼠无显著影响。BALB/c小鼠胎盘CD45+细胞群中TLR3表达率升高,并伴有CD45+CD80+细胞和CD8α+CD80+细胞百分率的升高。另可检测到CD45+细胞内IL-2表达率增高,而IL-10表达率下降;而在NOD/SCID模型中未观察到这种趋势。阻断TLR3能消除dsRNA对BALB/c小鼠胚胎排斥的诱导作用,并同时降低CD45+细胞群中CD80的表达率。结论:dsRNA与TLR3的相互作用可能参与CD45+CD80+细胞和CD8α+CD80+细胞的动员,随后母-胎界面IL-2表达增强,而IL-10表达减弱,最终导致胚胎被排斥。dsRNA引起的胚胎排斥过程可能需要具有正常功能的T细胞和NK细胞参与。
