ASC和p16基因启动子甲基化与非小细胞肺癌发生发展的关系 被引量:4

摘要 目的探讨ASC和p16基因启动子甲基化与非小细胞肺癌发生发展的关系。方法应用甲基化特异性PCR(MSP)检测非小细胞肺癌中ASC和p16基因启动子的甲基化发生率。结果48例非小细胞肺癌中ASC和p16基因启动子甲基化的发生率分别为47.9%和37.5%,显著高于癌旁正常组织(P<0.001);重度吸烟患者的肿瘤组织中ASC和p16基因启动子甲基化的发生率明显增高(P<0.05);ASC基因启动子甲基化在较小的肿瘤组织中的有较高的发生率(P<0.05)。而且,重度吸烟患者的正常肺组织中亦可检测到ASC基因启动子的甲基化。结论ASC和p16基因启动子的甲基化在非小细胞肺癌中有较高的发生率。ASC基因启动子的频繁甲基化可能与吸烟引起的非小细胞肺癌有关,并且ASC基因甲基化有可能是非小细胞肺癌发展过程中的一个早期事件。 [ Objective ] To investigate the correlation between methylation of ASC and p16 genes promoter region with the genesis and progress of NSCLC. [Methods] Methylation-specfic PCR (MSP) was used to detect methylation of ASC and p16 genes in promoter region. [Results] In 48 eases of NSCLC patients, there was a significant higher rate of ASC and p16 genes methylation in cancer tissues than in adjacent normal tissues (47.9% to 8.3%, 37.5% to 10.4% P 〈0.001); the frequence of methylation of ASC and p16 genes promoter was higher in the cancer tissues of heavy smoking patients (P 〈0.05) and in the smaller primary tumor tissues (P 〈0.05). Methylation of ASC gene promoter was detected in cancer adjacent normal tissues of heavy smoking patients. [Conclusions] There is a higher rate of ASC and p16 genes methylation in NSCLC; the frequency of methylation of ASC and p16 genes promoter may be related with NSCLC caused smoking and methylation of ASC gene may be an early event in the genesis and progress of NSCLC.
