摘要
以体外组织块培养的方法成功地建立了三个人眼小梁细胞系标本。在倒置显微镜下观察了三代小梁细胞的生长过程,用透射和扫描电子显微镜观察了第三代融合的小梁细胞的超微结构。小梁细胞的生长潜伏期约为1周,原代和传代融合的小梁细胞呈单层细胞生长,单个细胞呈长而扁平状,核位于中央为椭圆形,细胞表面大部分平滑,偶有微绒毛突出,在某些部位可见邻近细胞和其胞突间紧密连在一起。另外,这些细胞还具有高代谢细胞的超微结构特征包括大量的线粒体,发达的Golgi器,丰富的粗面内质网和明显的溶酶体。人小梁细胞体外培养为进一步研究小梁细胞的功能提供了非常有价值的实验物质基础,还可作为青光眼发病学和药物筛选等方面实验研究的基础。
Three trabecular cell lines were successfully established by means of tissue culturing in vitro.The growth patterns of the trabecular cell culture were studied under inverted microscope for 3 generations.The morphological features and ultrastructure of the third-passage confluent trabecular cells were investigated by transmission and scan electron microscopy.The latent period prior to growth was about 1 week.In primary confluent and subconfluent culture,the trabecular cells,grew in a monolayer.Individual cells had flattened,elongated cell bodies,a centrally located,oval nucleus,and generally smooth cell surface with occasional microvillous projections;in places,adjacent cells and their processes adhered closely one to another.In addition they had the altrastructural features of metabolically active cells including many mitochonctria,a well-developed Golgi apparatus,abundant rough endoplasmic reticulum and prominent lysosomes. Propagation of human trabecular cells in vitro may provide a valuable source of experimental material to study the functional aspects of these cells which line the trabecular meshwork.They may be also used in pathogenetical and pharmacological researches of glaucoma.
