双巯基乙酸异辛酯二正辛基锡对大鼠生殖内分泌激素的影响 被引量:1

摘要 目的研究双巯基乙酸异辛酯二正辛基锡(IMA)是否具有环境内分泌干扰物质样作用。方法18只受孕Wistar雌鼠随机分成6组,每组3只,产后每组留取体重相近的雌、雄幼鼠各5只,在出生24h内分别皮下注射0.1000、0.0100、0.0010、0.0001μg/10g(以玉米油为溶剂,相当于10.00、1.00、0.10和0.01μg/kg)的IMA(IMA实验组)和同等剂量的玉米油(阴性对照组)与0.001μg/10g(0.1μg/kg)的二月桂酸二丁基锡(DBTD,阳性对照组)。给药量为0.1ml/10g(10ml/kg),每天1次,连续5d。1.5个月后断头取血,检测血清睾酮(T)、促卵泡生成素(LH)、促黄体生成素(FSH),计算性器官指数。并做病理切片,观察性器官的微观结构改变情况。结果0.1000、0.0100μg/10gIMA组雄、雌仔鼠血清T浓度明显高于玉米油组,呈剂量-效应关系(雄鼠:r=0.950,P<0.05;雌鼠:r=0.911,P<0.05),组间差异有统计学意义(均P<0.01);各组雄、雌鼠血清LH、FSH浓度之间差异无统计学意义(均P>0.05);各IMA实验组雄鼠睾丸重量、睾丸指数均大于玉米油组(均P<0.05)。结论IMA对大鼠具有内分泌干扰作用,可引起Wistar大鼠血清中雄激素水平明显升高。 Objective To understand whether Imarcaptoacetate dioctyhin(IMA) is a environmental endocrine disruptor or not. Methods The new born Wistar rats were randomly divided into six groups including positive control group, negative control group and four experimental groups. The rats in the experimental groups were given IMA at different concentrations (0.100 0, 0.010 0, 0.001 0, 0.000 1μg/10 g) 1/1 000 000 by hypodermic injection, the rats in the negative control group were treated with corn oil and those in the positive control group were treated with dibutyhin dilaurate (DBTD) at concentration of 0.001μg/10 g bw. The rats were injected one time each day for five consecutive days. After one and half months, the levels of T, LH and FSH in the blood, the weight of the testicles, ovaries and the sexual organ exponent were determined. The pathological examination on some samples was conducted. Results In both of female and male, the levels of T in IMA treated rats were significantly increased and showed an doge-effect relationship (P〈0.01), as to the levels of LH or FSH there was no differences were seen among groups (P〉0.05). The exponents of the testicles were different among groups (P〈0.01), the negative group was the lowest (P〈0.05), the exponents of the ovaries were also different among groups (P〈0.05). Conclusion IMA has an effect on the sex hormone of rats and can be considered as an environmental endocrine disruptor.
