摘要
目的本文主要论证青霉素G间歇反萃取工艺的优点,对连续反萃取和间歇反萃取工艺原理进行了研究。方法将Bs与15%的K2CO3水溶液连续加入离萃取机,根据BA效价控制K2CO3的流速,使pH值保持在6.6~6.8之间进行反萃机。分离得取RK送到结晶工序。结果通过研究分析和实践证明。间歇反萃取较连续反萃取能够更好的满足结晶对PK的质量要求,即pH值更稳定,杂质含量低,水份含量低。因此,结晶时间明显缩短。青霉素钾盐的质量和吸收率显著提高。结论通过以上的研究分析和论证,我们得出青霉素G间歇反萃取工艺的优越性在于:能使二氧化碳彻底排出,加快反应速度,缩短操作时间,减少青霉素的损失。以分层分离的方式代替离心分离,避免了气密现象的发生。
Objective This article has mainly demonstrated the advantages of discontinuous reverse - extraction process of Penicillin G,and investigated the process principle of consecutive reverse extraction and dicontinuous reverse - extraction and analyzed the factors that influence the quality and yield of reverse - extraction products. The article has demomtrated the superiority of discontinuous reverse - extraction process of Penicillin G by comparing the related data of consecutive reverse extraction with those of discontinuous reverse extraction. We also compared the related data of PK obtained from consecutive reverse - extraction and discontinuous reverse - extraction in the crystallization process and demonstrated the superiority of discontinuous reverse extraction process of Penicillin G agagin. Methods Add BA and 15% of K2CO3 aqueous solution to centrifugal extractor continuously; control the flow rate of K2CO3 according to the potency of BA. Keep the pH to 6.6 - 6.8. Perform reverse extraction. Obtain PK by separating and then send to crystallization process. Results Through analysis and practice, it proves that discontinuous reverse extraction can satisfy the quality requirements of PK better than consecutive reverse extraction, i. e. pH is more stability, the content of impurities are lower, and water content is lower. So crystallization time is obviously shorter. Conclusion Thi-ough the above the analysis, we can conclude that the advantage of discontinuous reverse- extraction Penicillin G is that CO2 exhaust completely, and that the reaction speed is rapid,and the operation time is short, and the lass of product is low. Meanwhile,this method can avoid the occurring of airtight because the layer separationreplaces the centrifugal separation.
