大豆异黄酮β-葡萄糖苷酶产生菌的选育及产酶条件研究 被引量:4

摘要 以豆豉为材料筛选产大豆异黄酮-β葡萄糖苷酶的菌株为出发菌,通过UV和LiCl诱变育种,利用单因素和正交试验进行产酶条件研究,确定最佳工艺条件为:黄豆粉1%,酵母膏1%,麸皮2%,KH2PO40.1%,NaCl0.5%,起始pH值7.5,装液量30mL/300mL,发酵时间48h,发酵温度37℃,转速70r/min。测得酶活为5.49U/mL。 Soybean isoflavone β-glucosidase-producing strain was screened from lobster sauces. The strain with the highest deniposide enzyme activity was used. The original strain was identified as bacillus by microscope inspection. The initial strain was mutagenized with ultraviolet radiation (UV) and LiCl. The optimum enzyme-producing conditions were obtained by single factor and orthogonal experiments: (v/v or w/w) soybean flour 1%, yeast extract 1%, bran 2%, KH2PO4 0.1%, NaCl 0.5%, original pH 7.5, liquid load 30 mL/300 mL, fermentation time 48 h, fermentation temperature 37℃, rotation speed 70r/min. Under the optimal conditions, the deniposide enzyme activity was 0.826 and the salicin enzyme activity was 5.49 μg/mL.
