两种纯化玉米超氧化物歧化酶方法的比较 被引量:2

Comparison of Two Procedures for Purifying SOD from Maize

摘要 本文利用硫酸铵分级沉淀、凝胶过滤初步分离了玉米SOD,利用DEAE-52离子交换层析和金属螯合亲和层析分别纯化了SOD。其中经离子交换层析后的比活为3 304,纯化倍数为118,回收率为4.8%,基本达到电泳纯。经金属螯合亲和层析纯化后比活为8 597,纯化倍数为307,回收率为29.4%,达到了电泳纯。 Crude enzyme Superoxide dismutase (SOD) was obtained from maize by precipitation with ammonium sulfate and sephadexG - 100 ; then the SOD was purified by two methods, DEAE - 52 chromatography and metal chelating affinity chromatography (MCAC) separately. As showing in the result, the specific activity of DEAE - 52 purified SOD is 3304 with a purification factor of 118, and the specific activity of MCAC purified SOD is 8591 with a purification factor of 307. It is found that the corn SODs purified by both methods are homogeneous and their MW are all 1.6KD in SDS- PAGE.
