摘要 目的:测定通脉四逆汤中乌头类生物碱的煎出量及变化规律.方法:采用RP-HPLC法,Hypersil-BDS柱(250mm×4.6mm,5μm),以甲醇-水-氯仿-三乙胺(100:50:3:0.15)为流动相,流速为1mL·min^-1,波长为234nm,柱温:30℃.结果:乌头碱、中乌头碱和次乌头碱在(0.0025~0.0800)μg、(0.0028~0.0880)μg和(0.0023~0.0760)μg范围内线性关系良好,r分别为0.9997、0.9996、0.9996.结论:该方在煎煮过程中,同煎的其他药物对乌头类生物碱在汤中的含量产生影响,文火滚煎约70min后,药汤中乌头碱和次乌头碱成分基本消失,中乌头碱消失过半. Objective :To measure the contents of aconitine alkaloids in Tongmai-Sini-Tang, and its regularity of vary with decoction time. Methods: A RP-HPLC method was used. The chromatographic conditions are listed below: Hypersil-BDS column(250mm×4.6mm,5μm),a mobile phase composed of methanol-water-chloroform-triethylamine( 100 :50:3 : 0.15), a flow rate of 1 mL·min^-1 and wavelenghts-234nm, column temperature was 30℃. Results: The linear range of aconitine was (0.0025 - 0.0800) μg ( r = 0. 9997), mesaconitine was (0.0028 - 0.0880) μg ( r = 0.9996), and hypaconitine was (0.0023- 0.0760)μg( r = 0.9996). Conclusion: while decocting medicinal herbs except from major herb in the formulation, other herbs were effective on the contents of aconitine alkaloids, the compositions of aconitine and hypaconitine would disappear after decocting medicinal herbs for 70 minutes, but the mesaconitine disappear only half.
