摘要
目的:探讨皖南蝮蛇蛇毒抗凝组份对白血病细胞HL60和K562增殖的抑制作用。方法:采用细胞形态学观察,台盼蓝染色,MTT比色法测定蛇毒抗凝组份Ⅱ和Ⅲ(ACFⅡ和Ⅲ)对体外培养的白血病细胞株HL60和K562增殖的抑制作用。结果:蛇毒ACFⅢ对白血病细胞HL60和K562均有较强的抑制作用,蛇毒ACFⅡ对白血病细胞HL60和K562的影响经统计学分析无明显统计学意义。蛇毒ACFⅢ对HL60和K562的抑制作用呈剂量时间依赖性。结论:蛇毒ACFⅢ对白血病细胞系HL60和K562细胞的增殖有明显的抑制增殖和诱导凋亡的作用。
Objective:To explore the inhibitory effect of anticoagulant fraction from Agkistrodon(found in South Anhui Province) halys pallas venom on the ceil proliferation in HL-60 cells and K562 ceils. Methods : Cell morphological transformation was observed with inverted microscope, and cell proliferation was tested by MTT assay and trypan blue staining. Results:ACFⅢ peak showed obvious inhibitory effect on the cell proliferation of HL-60 and K562 cells and the inhibitory effect correlated with time and dosage. However, no inhibitory effect on the cell proliferation of HL=60 and K562 cells was observed from ACF Ⅱ peak by statistic analysis. Conclusion :Agkistrodon halys pallas venom ACF Ⅲ can inhibit and kill HL 60 and K562 cells.
