PAS结构域介导Org35蛋白与NifA之间相互作用 被引量:1

摘要 在巴西固氮螺菌中,氧和铵是调节固氮基因表达的主要环境因子,即高浓度的氧和铵抑制固氮作用.NifA蛋白(nifA基因的产物)是所有固氮基因(nif基因)的转录激活蛋白。目前,在该菌中,氧和铵对NifA蛋白的表达和活性的调节机制仍然不清楚.本研究从巴西固氮螺菌中克隆到编码含有PAS结构域蛋白的org35全基因,将根据org35的核苷酸序列推测的Org35蛋白氨基酸序列在NCBI网上进行同源性比较,结果表明,Org35蛋白属于杂合双组分调节系统,包括3个结构域,即N-端PAS结构、中间组氨酸激酶结构域(HPK)和C-端响应调节蛋白结构域(RR).并通过酵母双杂交系统证明了Org35蛋白与NifA蛋白之间的相互作用是由PAS结构域介导的.
