在2.2.15细胞中siRNA对HBV复制和表达的抑制

siRNA mediated inhibition of HBV replication and expression in 2.2.15 cells

目的:检测设计的siRNA对HBV复制和表达的影响。方法:选择针对乙型肝炎病毒(HBV)的siRNA基因序列,化学修饰合成3种stealth siRNA,脂质体转染带有HBV的2.2.15细胞,ELISA法及荧光定量PCR检测其对HBV复制和表达的影响。结果:3种stealth siRNA对HBV的复制和表达在不同时间有不同程度的抑制作用。结论:HBV在2.2.15细胞中的复制和表达被stealth siRNA抑制。

Objective： To study the effect of siRNA on HBV replication and express：ion. Methods： The siRNA against HBV were designed and synthesized, then the stealth siRNAs were synthesized by modification of siRNA. Lipofectamine^TM 2000 reagent was used for the transfection of 2.2.15 cells by stealth_508, stealth 2311 and stealth_2765 or control. Concentration of HBsAg and HBeAg in the supematant of the transfected cells were measured using ELISA. Copies of HBV in the transfected cells was measured using quantitative PCR. Results： Compared with controls, these stealth siRNA against HBV decreased concentration of HBsAg and HBeAg and copies of HBV significantly. Conclusion： HBV replication and expression in 2.2.15 cells are inhibited by stealth siRNA.
