农杆菌介导白细胞介素-2基因转化大白菜的研究 被引量:2

摘要 以大白菜4 ̄5d苗龄、带2mm子叶柄的子叶为外植体,利用农杆菌介导的方法将人源白细胞介素-2基因(il-2)导入北方大白菜优良品种鲁白七号和602中,分别获得带有PPT抗性和卡那霉素抗性的两种转化植株,转化率分别最高达到16.7%和8.0%,讨论分析了两种质粒(PPT和卡那霉素两种抗性标记基因)对转化效率的影响。转化植株进行PCR和PCR-Southern杂交、Western杂交检测,结果证明白细胞介素-2基因已整合入白菜基因组中,il-2蛋白在转基因大白菜中得到表达。 The 5days old of cotyledons with 2mm petiole were used to be as explants, interleukin-2 gene (il-2)were transferred into Chinese cabbage by Agrobaeterium-medieted method.The transformed plants with the resistance to PPT and Kanamiein were gained.By the PCR assay,the frequency of transformed Chinese cabbage reached 16.7% and 8.0% respectively. Then some of them were examed by PCR-Southern bloting and Western bloting.So the integration of il-2 into Chinese cabbage genome and the expression of il-2 in transformed plants were verified.
