摘要
云锦杜鹃是具有较高观赏价值和利用价值的野生珍稀植物。以新发茎段为外植体,以WPM为基本培养基,研究了云锦杜鹃的组培快繁技术。结果表明,附加外源激素1 ̄5mg/L浓度的ZT能诱导腋芽分化,6 ̄8周继代一次。不同浓度ZT对继代培养中的再生枝条数有显著影响,以0.5 ̄1mg/LZT处理的增殖系数和组培苗生长为优;不同pH值对增殖系数没有显著影响,但对组培苗长势有明显作用,以4.8 ̄5.2处理较为适宜。将组培苗扦插在无土基质中,达到85%以上的瓶外生根率。
Rhododendron fortunei is a wild ornamental plant with highly appreciative and useful value. In vitro micropropagation technology of Rhododendron fortunei was aimed to be established using shoots as explants and WPM as fundamental culture medium. The results indicated that axillary shoots could be induced by the medium supplemented with a concentration of 1-5 mg/L zeatin, and it needed 6-8 weeks for one regeneration. Different concentration of zeatin had a significant influence on regenerated shoots. The best hormone concentration for shoot regeneration was at 0.5-1 mg/L from quantitative and qualitative views. The pH value which had obvious effect on shoot growth was appropriate at 4.8-5.2, although showing no significant difference in regenerated shoots. In vitro rooting percentage of 85% was obtained when unrooted shoots were directly transplanted into soilless mixtures.
