摘要
目的:探讨雌激素对人精子胞浆内游离钙离子浓度([Ca2+]i)的影响及其与精子功能状态之间的关系。方法:选择18例生育力正常男性精液标本,其中10例制备获能精子,8例制备非获能精子。将获能精子和非获能精子分别用不同浓度的17β-雌二醇(E2)作用,之后以流式细胞术检测作用前后精子内荧光强度值的变化以反应E2对[Ca2+]i的影响。结果:一定浓度的17β-雌二醇可引起正常人获能精子[Ca2+]i显著升高,17β-雌二醇对非获能精子[Ca2+]i无明显影响。结论:雌激素可通过诱导[Ca2+]i升高而激活人精子,该激活作用可能与精子的功能状态有关。
Objective: To investigate the effect of estrogen on intracellular calcium concentration ( [ Ca^2+ ] i ) in human spermatozoa and the possible relationship between changes of [ Ca^2+ ]i and the functional states of spermatozoa. Methods: eighteen samples of normal fertile men were selected. Ten were prepared for capacitated spermatozoa and eighteen for uncapacitated spermatozoa. Both capacitated and uncapacitated spermatozoa were stimulated by 17β-estradiol( E2 )at different concentrations. Changes of [ Ca^2+ ]i were detected by flow cytometry and the fluorescence intensity was measured to evaluate the [ Ca^2 +] i. Results: The data showed that E2 at appropriate concentrations sig- nificantly increased the [ Ca^2 + ]i in capacitated spermatozoa, while E2 had no effects on [ Ca^2 + ]i in uncapacitated spermatozoa. Conclusion: Estrogen can activate human spermatozoa by rapidly increasing the [ Ca^2 + ] i and this role of estrogen is perhaps associated with the functional states of spermatozoa.
