摘要
目的研究盐酸坦洛新对映体在纤维素衍生物及蛋白质固定相上的保留行为,优化色谱条件。方法采用Chiralcel OJ,Chiral- cel OD-RH和Chiral AGP为固定相,流动相分别为正己烷-乙醇-二乙胺(70:30:0.1)、50 mmol·L^(-1)六氟磷酸钾溶液(pH 5.8)-乙腈(65:35)和10 mmol·L^(-1)磷酸盐缓冲液(pH 5.8)-乙腈(93:7);检测波长均为225 nm,并考察了影响分离的因素。结果盐酸坦洛新对映体在上述3种手性固定相均达到基线分离(R_s>1.5)。结论采用上述3种固定相均可方便地分离盐酸坦洛新对映体。
OBJECTIVE To develop a HPLC method for the separation of the enantiomers of tamsulosin hydrochloride using derivatized cellulose and protein chiral stationary phases. METHODS The separations were performed on Chiralcel OJ, Chiralcel OD-RH and Chiral AGP with the mobile phase of hexane-alcohol-diethyl-amine (70 : 30 : 0.1 ), 50 mmol·L^-1 potassium Hexatluorophosphate solution( pH 5.8 )- acetonitrile(65:35) and 10 mmol·L^-1 phosphate buffer solution (pH 5.8)-aeetonitrile(93:7), respectively. The detective wavelength is 225 nm. Several parameters on separation were studied. RESULTS Baseline separations( Rs 〉 1.5) were easily obtained in all cases. CONCLUSION The enantioseoparation of tamsulosin hydroctdoride was achieved conveniently with three types of the CSPs used in this study.
