摘要
目的:建立测定人血浆中去羟肌苷浓度的反相高效液相色谱方法。方法:血浆中加入内标后经固相萃取,色谱柱为 A-pollo C_(18)(150 mm×4.6 mm,5 μm),流动相:乙腈-水(8:92),流速:0.9 mL·min^(-1),检测波长250 nm。结果:本方法线性检测范围为0.025~10 μg·mL^(-1),r=0.9997,定量限为0.025 μg·mL^(-1),绝对回收率为72.49%~77.09%,相对回收率为99.54%~101.5%,日内、日间精密度的 RSD 均小于6%。结论:本方法灵敏度高,操作简便,可用于人血浆中去羟肌苷的浓度测定及临床药代动力学研究。
Objective:To establish an HPLC method for the determination of didanosine in human plasma. Methods:Separation was achieved on a Apollo C18 column( 150 mm ×4. 6 mm ,5 μm) and detected at 250 nm. The mobile phase consisted of acetonitrile and water(8: 92)at a flow rate of 0. 9 mL · min^-1. The sample was prepared by solid phase extraction. Results :The linear range was 0. 025 - 10 μg·min^-1, r = 0. 9997, the limit of quantification was 0. 025 μg·min^-1. Absolute recovery was 72.49% -77.09% ,and relative recovery was 99. 54% - 101.5% ; intra- day RSD and inter- day RSD were less than 6%. Conclusion:The method is sensitive,convenient, and is suitable for the determination of didanosine concentration in human plasma and pharmacokinetic studies.
