氨基胍和维生素C调节1型糖尿病大鼠胰腺脂肪细胞膜iNOS的表达

摘要 目的观察氨基胍、维生素C对糖尿病大鼠胰腺的脂肪细胞膜iNOS蛋白表达的影响。方法利用链脲佐菌素腹腔注射法诱导建立1型糖尿病大鼠模型,SD大鼠随机分为正常对照组、糖尿病组、维生素C治疗组、氨基胍治疗组,治疗16周后取胰腺组织作免疫组织化学检测胰腺腺细胞的脂肪细胞膜iNOS蛋白表达情况。结果(1)糖尿病组胰腺脂肪细胞膜iNOS阳性产物呈弥漫分布,其蛋白表达明显高于正常对照组;(2)氨基胍、维生素C组iNOS蛋白表达明显低于糖尿病组。结论1型糖尿病大鼠胰腺的脂肪细胞膜iNOS蛋白表达升高,氨基胍(AG)、维生素C(VC)则对糖尿病组大鼠脂肪细胞膜i-NOS蛋白质表达具有抑制作用。 Objective To investigate the effect of aminogucinidine and vitaminC on the expression of iNOS protein of adipose cell membrane in diabetic rat pancreas. Methods Diabetic rats were induced by intraperitoneal injection of STZ. Rats were randomly divided into four groups: normal control; diabetic model; vitaminC administmtion; aminogucinidine administration. After being fed for 16 weeks,the rots were killed,and the pancreas were separated to observe the expression of iNOS protein of adipose cell by the method of i mmunohistoehemieology. Resuits ( 1 ) The protein expression of iNOS on adipose cell membrane in diabetic group was higher than that of nornal control group ( P 〈 0.05 ). (2) The expression of iNOS protein in both of aminogueinidine and vitaminC group were lower than that of diabetic group. Conclusion Both of aminogueinidine and vitaminC could inhibit the expression of iNOS protein on adipose cell membrane in type Ⅰ diabetic rat pancreas.
