大肠癌并肝转移的蛋白质组学研究

摘要 目的采用蛋白质组学技术研究有转移组和无转移组大肠癌组织蛋白质组表达的差异,分析和鉴定两组间特异表达的肿瘤蛋白质,探索大肠癌并肝转移发生的机制。方法用等电聚焦/SDS聚丙烯酰胺双向凝胶电泳对比分析20例大肠癌患者原发灶、癌旁肠黏膜和肝转移灶中蛋白表达,经肽质量指纹谱分析、鉴定差异蛋白质。结果双向电泳图像对比分析表明,癌旁大肠黏膜组织、癌原发灶和肝转移灶中蛋白质表达有明显差异。在肝转移组中存在AnnexinA2蛋白和JE0350蛋白特异表达,磷酸丙酮酸水合酶和CAC15744蛋白表达上调;在无肝转移组存在CAD80231和A38983蛋白特异表达,AAH01166蛋白表达上调。结论大肠癌并肝转移者与无肝转移者在蛋白质组表达上存在一定的差异。对这些差异蛋白质的进一步深入研究,可帮助了解大肠癌易发生远处转移的蛋白质组学机制及采取相应的处理对策。 Objective To analyze and identify the differentially expressed between two groups,one with hepatic metastasis and the other without hepati proteins of colorectal carcinoma tissues c matastasis and to study the mechanism of hepatic matastasis in colorectal carcinoma by using two- dimensional gel electropresis and mass spectrum analysis. Methods Isoelectric focusing/SDS acrylamide gel two- dimensional electrophoresis was used to analyze differential expression protein among normal colorectal mucosa , primary cancer lesion and hepatic metastasis. Peptide mass fingerprinting was used to identify the different proteins. Results Significant differences of protein expression were found on two - dimensional electrophoresis. We detected JE0350 and Annexin A2 protein expressed exclusively and phosphopyruvate hydratase alpha and CAC15744- like protein increased in the carcinoma group with hepatic metastasis. We also detected A38983 and CAD80231 - like protein expressed exclusively and AAH01166 protein increased in the carcinoma group without hepatic metastasis. pressed proteins existed in the two cancer groups.
