摘要
目的:探索组织块细胞培养方法构建子宫内膜异位症(内异症)腹膜红色病变的细胞体外模型.方法:采用组织块培养法共培养12例腹膜红色病灶,同时以酶消化共培养法对2例在位子宫内膜进行细胞培养作为实验对照.进行抗角蛋白和波形蛋白以及抗Flt-1、KDR抗体的细胞免疫化学检测.结果:12份标本中8份培养成功,每75ml体积的培养瓶可得到(2~5)×10^6个腺细胞及间质细胞,与酶消化培养法子宫内膜组织细胞的形态相似,经细胞免疫化学验证细胞成分并进行了初步细胞学实验研究.结论:腹膜红色病变组织的组织块培养法可作为内异症早期病变的细胞体外模型之一.
Objective:To explore the tissue mass cell culture of peritoneal red lesions as in vitro cell model of endometriosis. Methods:The tissue mass cell culture was used in the twelve samples of peritoneal red lesions with the endometriosis. The enzymolysis cell culture was used to the 2 samples of endometrium with endometriosis for the control group. The immunocytochemistry of anti-cytokeratin antibody, anti-vimentin antibody, anti-Fh-1 receptor antibody and anti-KDR receptor antibody were used for the cells. Results:The cell culture was successfully established in 8 of 12 peritoneal red lesions. From one 75ml cell culture bottle,the yield of primary culture of stroma cells and glandular epithelial cells were about(2~5)×10^6. The morphology of the red lesions cell was the same as the endometrium of endometriosis and confirmed by immunocytochemistry. The cells used for the primary cell investigation. Conclusion: The tissue mass cell culture of peritoneal red lesions can be used as in vitro cell model of early stage endometriosis.
