摘要
目的探讨信号传导和转录活化子(activatedsignaltransducerandactivatoroftranscription3,Stat3)和血管内皮生长因子(vascularendothelialgrowthfactor,VEGF)在人胃癌细胞中表达及其与临床病理特征的相关性。方法利用荧光定量聚合酶链反应法测定32例人胃癌细胞和25例癌旁细胞内Stat3mRNA和VEGFmRNA的定量表达,分析这些指标的相互关系及其与临床病理特征的相关性。结果人胃癌细胞中Stat3mRNA和VEGFmRNA的表达与性别、年龄无关(P>0.05)。人胃癌细胞中Stat3mRNA和VEGFmRNA的表达明显高于癌旁细胞的表达(P<0.01)。人胃癌细胞中Stat3mRNA和VEGFmRNA的表达与组织类别、浸润深度、分化程度、淋巴结转移、分期和大小有关(P<0.05)。人胃癌细胞中Stat3mRNA与VEGFmRNA呈显著正相关(r=0.63303,P=0.0001)。结论Stat3mRNA和VEGFmRNA表达上调与人胃癌发生、发展和转移有关,JAK/Stat信号传导途径可能与VEGFmRNA在胃癌细胞过度表达有关。
Objective To determine expression of activated signal tra. ducer and activator of transcription 3 and vascular endothelial growth factor,and its relation to clinic pathological characteristics in human gastric cancer. Methods Expression of Star3 mRNA and VEGF rnRNA in 32 cases gastric cancer and their adjacent noncancerous tissues was measured by fluorescent quantitative polymerase chain reaction (FQ-PCR). Results Expression of Star3 mRNA and VEGF mRNA was not correlated with sex, age in human gastric cancer (P〉0. 05), Expression of Stat3 mRNA and VEGF mRNA in human gastric cancer was significantly higher than that in their adjacent noncancerous tissues (P〈 0. 01). Expression of Stat3 mRNA and VEGF mRNA was correlated with histological grades,depth ot7 tumor invasion,tumor differentiation,lymph node metastasis and tumor size in human gastric cancer (P 〈0.05). Marked correlation in human gastric cancer were found between expression of Star3 mRNA aud VEGF mRNA (r = 0. 633 03,P = 0.000 1), Conclusions Our results show that over expression of Star3 mRNA and VEGF mRNA could play an important role in the development, process and metastasis of human gastric cancer, Deregulated JAK/STAT signaling pathway may play an important role in VEGF over expression in gastric cancer and contribute to gastric cancer development and progression.
