催产素与其反义肽抗体的相似性

SIMILARITY BETWEEN THE OXYTOCIN AND A ANTIBODY OF ANTISENSE PEPTIDE OXYTOCIN

观察了催产素反义肽抗体（ａｎｔｉｂｏｄｙｏｆａｎｔｉｓｅｎｓｅｐｅｐｔｉｄｅｏｘｙｔｏｃｉｎ，简称ＡＳ－ ＯＴ－Ａｂ）对催产素受体（ＯＴＲ）的识别特性。该反义肽是按照牛催产素（ＯＴ）和它的结合蛋白ＮｅｕｒｏｐｈｙｓｉｎＩ（ＮＰＩ）氨基端前十二个氨基酸（包括ＯＴ［１－９］）编码ＤＮＡ的互补序列固相合成的，简称ＡＳ－ＯＴ。ＡＳ－ＯＴ与甲状腺球蛋白联结后，制备兔抗ＡＳ－ＯＴ多克隆抗体（ＡＳ－ＯＴ－Ａｂ），间接酶联免疫吸附测定（ＥＬＩＳＡ）证明，ＡＳ－ＯＴ－Ａｂ能与催产素受体（ＯＴＲ）相互结合，而这一结合能被ＯＴ所阻断，显示了ＯＴ与其反义肽抗体的相似性。

We observed the interaction properties of the antibody of antisense peptide oxytocin whose sequence is encoded in antisense strand of DNA corresponding to the 12 amino-terminal residures of bovine oxytocin and its binding protein neurophysin I(NPI),a region that included the OT sequence at residues l-9.The antibody was shown to bind selectively to oxytocin receptor in plasma membrane from rat mammary gland after delivery by ELISA.The binding was inhibited by oxytocin.It was shown the similarity between the oxytocin and a antibody of antisense peptide oxytocin.
