局灶性缺血再灌注时丹参对热休克蛋白70的影响-免疫细胞化学及病理学研究 被引量:10

摘要 丹参素以活血化瘀称著,常用于治疗缺血性脑血管病,热休克蛋白70(HSP70)具神经保护作用。本文研究目的为:丹参是否也通过影响 HSP_(70)起作用。采用大鼠犬脑中动脉(MCA)线检模型。以免疫细胞化学及病理组织方法在同一动物中进行研究。结果发现在缺血90min 再灌注24h 后.对照组手术侧皮层 HSP_(70)免疫反应增强及皮层神经细胞有缺血性损伤,而丹参组(缺血90min 时给予丹参10g/Kg,体重 ip),则 HSP_(70)免疫反应增强,不仅免疫反应阳性细胞在数量上,而且在染色强度上均显著比对照组明显,而相应皮层神经细胞的缺血性损伤则较轻。提示丹参对局灶性缺血再灌注损伤的保护作用,可能与它能影响热休克蛋白70合成机制有关。 Radix Salviae Miltorrhizae (RSM)has huoxue huaxue effect and is used to treat is- chemic cerebral vascular diseas.es.Heat shock protien 70 (HSP)has been proved to have neuroprotec- rive effect.The aim of the present study is to examine whether RSM has the effect on HSP_(70)in focal isehemic reperfusion rats using immunocytochemistry and histopathology methods in the same ani- mals.A middle cerebral artery thread embolism rat model was used.In immunocytochemistry study using anti—HSP 70 antibody,staining of cortical neurons was only slightly detected in sham—operat- ed animals.Strong staining was denonstrated in control animals after 90—min ischemia and 24 h reperfusion that in the contrel animals and the cortical neuronal ischemic damage was also much milder.The results suggest that RSM affords neuroprotection against focal ischemic reperfusion in- jury by the induction of HSP_(70).
