摘要
目的制备树突状细胞(dendritic cells,DC)与胶质瘤融合细胞,探讨其特异性抗肿瘤活性。方法诱导正常人外周血来源DC,在PEG作用下将其与胶质瘤细胞BT325融合,通过双荧光标记法检测融合率、MTT法测定CTL活性。结果融合细胞在培养体系中占较高比例;DC/胶质瘤融合细胞激活的CTLs对胶质瘤的杀伤作用显著高于DC与胶质瘤细胞联合培养DC组(P<0.05)及其他对照组,而且杀伤活性随着效靶比的增加而增加。结论DC与胶质瘤细胞融合是一种更为有效的以DC为基础的抗肿瘤策略。
Objective To prepare the fusion cells of dendritic cells and glioma cell line and evaluate their activity of antitumor. Methods DCs derived from normal peripheral blood are induced and they are fused with glioma cells under PEG. The fusion ratio is determined by two - colour fluorescent labeling. Cytotoxicity is determined by MTr method. Results The percentage of fusion cells in this culture system was high. The CTL activity against glioma cells induced by DC/glioma fusion cells was significantly higher than that induced by coculture of DC and glioma fusion cells as well as other control groups. The Cytotoxicity was increased as the ratio of effectors and target cells increased. Conclusion The fusion of DC and glioma cells is a more effective antitumor strategy among those based on DC.
