摘要
目的比较骨关节炎滑膜成纤维细胞与皮肤成纤维细胞基因表达的差异,以进一步研究滑膜成纤维细胞在骨关节炎中的作用。方法选取骨关节炎患者滑膜,经原代培养获得滑膜成纤维细胞,采用正常志愿者原代培养皮肤成纤维细胞作为对照。使用限制性酶切片段差异展示技术分离滑膜成纤维细胞与皮肤成纤维细胞差异表达之基因。结果骨关节炎患者滑膜成纤维细胞与正常志愿者皮肤成纤维细胞相比,SOD、TFPI2、CXCL2、CXCL6、TGF-β基因表达水平较高。结论SOD、TFPI2、CXCL2、CXCL6、TGF-β基因在骨关节炎滑膜成纤维细胞表达水平高于皮肤成纤维细胞中相应基因的表达水平,表明这些基因在骨关节炎疾病的发生过程中可能起到一定的作用,为进一步研究骨关节炎以及治疗骨关节炎提供了新的对象。
Objective Compare the difference in genes expressions of osteoarthritis synovial fibroblasts and skin fibroblasts so as to study the role of synovial fibroblasts in osteoarthritis. Methods Get the primary culture of synovial fibroblasts from patients of osteoarthritis. Primary culture skin fibroblasts from normal volunteer were used as control. Analyze the differences in the genes expressions of the two types of the cells using restriction fragment difference display method.Results The expressions of genes of SOD, TFPI2, CXCL2k, CXCL6 and TGF-β were higher in osteoarthritis synovial fibroblasts than those in normal skin fibroblasts. Conclusion The expressions of genes of SOD, TFPI2, CXCL2, CXCL6 and TGF-β were higher in the synovial fibroblasts showed that these genes play some certain roles in the development of osteoarthritis. These provide new targets for the study and the development of new treatment of osteoarthritis in future.
