摘要
目的探讨妊娠早期Granulated Metrial Gland(GMG)细胞对血管增生及胎盘形成的作用。方法对非妊娠及妊娠第5~17天小鼠的子宫标本分别行PAS试剂,抗血管内膜生长因子(VEGF)抗体及抗F-Ⅷ抗体染色,以测定不同时期胎盘组织的面积,其内血管面积及GMG细胞数。通过ELISA和RT-PCR方法测定妊娠期子宫内膜内VEGF的量变。利用双重染色,电镜及免疫电镜方法对产生VEGF的细胞进行鉴定。结果胎盘组织面积在妊娠第8~14天明显增大,妊娠第13天达到高峰;胎盘内血管面积于妊娠第7天起增大,妊娠第13天达到高峰,其与胎盘面积及胎盘内GMG细胞数增加的动态曲线相吻合。胎盘组织内VEGF含量于妊娠第7~11天达到高峰,于第13天起递减。结论GMG细胞可以产生VEGF,因此,对妊娠期子宫内膜内血管的增生以及胎盘组织的发育起着非常重要的作用。
Objective TO study function of GMG cells on placental development and angiogenesis. Methods No-pregnant mice with random cycling and pregnant mice at days 5 - 17 were used for experiments, The specimens were stained with PAS reagent, anti- VEGF antibody, and anti-F-Ⅷ antibody, We measured the total area of placenta and neovascularization. The content of VEGF was measured by enzyme-linked immunosorbent assay (ELISA). VEGF mRNA expression in the uterus was analyzed by the reverse transcription/ polymerase chain reaction (RT-PCR).Results The total area of the placenta per uterine section showed high values between day 8 and day 14, with peak at day 13, The total vascular area per uterine section started to increase at day 7, reaching a peak at day 13, in parallel with the increase of the placenta area and number of GMG cells. The content of VEGF was significandy elevated at days 7 - 11 and was slightly decreased at day 13. VEGF mRNA was expressed between day 7 and day 13, Conclusion These data indicate that GMG cells express VEGF and may play inducing roles in uterine neovascularization and improve placental development during pregnancy.
