摘要
目的探讨蛋白酪氨酸磷酸酶SHP-1 mRNA在γ射线体外诱发的白血病小鼠肿瘤细胞中的变化情况,为进一步从细胞信号转导途径入手研究辐射致癌的分子机制奠定一定的基础。方法实验分为癌变组、未癌变组及对照组,应用荧光定量PCR法分别检测各组胸腺及骨髓细胞中SHP-1 mRNA的含量变化情况。结果癌变组胸腺细胞中SHP-1 mRNA的含量与未癌变组及对照组比无明显差异,而骨髓细胞中SHP-1 mRNA的含量明显高于对照组。结论在γ射线诱发的白血病小鼠,胸腺细胞SHP-1 mRNA表达无明显变化,但存在SHP-1的翻译异常,表现为翻译增强现象,骨髓细胞中SHP-1 mRNA的表达明显增强。
Objective To investigate the alteration of SHP-1 mRNA of tumor cells of mice with γ ray-induced leukemia. Methods BALB/c mice were randomly divided into eaneeration group, non-caneeration group and control group. The content of SHP-1 mRNA in thymus cells and bone marrow cells were detected by real time fluorescence quantitative PCR(FQ-PCR). Results No significant difference in change of SHP-1 mRNA content was found in thymus cells in canceration, non-canceration and control groups. The SHP-1 mRNA content of bone marrow cells in eaneeration group was significantly more than that in controls. Conclusion There is translation abnormality of SHP-1 mRNA in thymus cells of mice with γ ray-induced leukemia, which is presented as translational enhancement. There is expression enhancement of SHP-1 mRNA in bone marrow cells of mice with γ ray-induced leukemia.
