EGF和TGF-α对多囊卵巢综合征患者颗粒细胞分泌E_2的影响 被引量:1

Action of EGF and TGF α on oestradiol production by granulose cells from patients with polycystic ovary syndrome

摘要 目的:探讨表皮生长因子(EGF)和转化生长因子-α(TGF-α)对离体多囊卵巢综合征(PCOS)患者卵巢黄素化颗粒细胞分泌雌二醇(E2)的影响。方法:收集体外授精胚胎移植(IVF ET)时PCOS组和正常组的黄素化颗粒细胞进行体外原代培养,在其培养的不同时间(0h、48h、120h)以睾酮(10-7mol/L)为底物,分别添加EGF(10μg/L)和/或TGF-α(10μg/L)于无血清培养液(TCM199)中,共培养3h后收集培养细胞。用放射免疫法(RIA)测定颗粒细胞分泌E2量;用考马斯亮蓝G250/BSA测颗粒细胞蛋白含量,以校正E2含量。结果:与对照组相比,在0h,EGF,TNF-α或EGF+TGF-α作用的PCOS组E2的含量均明显升高(P<0.05);而在48h、120h,E2含量均明显降低(P<0.05)。结论:体外培养的PCOS患者颗粒细胞有较高的芳香化酶活性;EGF、TGFα可显著抑制PCOS患者颗粒细胞E2的分泌。 Aim : To investigate the effects of EGF and TGF-α on oestradiol production by the ovarian luteinized granulosa cells from polycystic ovary syndrome (PCOS) patients. Methods:The luteinized granulosa cells were prepared from follicular pump, and pre-incubated for 0 h, 48 h, 120 h, then the granulosa cells were cultured with EGF ( 10 μg/L) , TNF-α (10μg/L) and EGF + TNF-α for 3 h in the presence of testosterone as substrate. Finally, the cells were collected for the measurements of oestradiol by RIA, and their protein concentration was assayed by radioimmunity. Results : At 0h in PCOS group, the addition of EGF and/or TGF-α to the cultures induced a significant increase in oestradiol production than those of normal group ( P 〈 0. 05 ) ; but for 48 h, 120 h in PCOS group, the addition of EGF and/or TGF-α to the cultures induced a significant decrease in oestradiol production than those of normal group (P 〈0.05) . Conclusion: Granulose cells from PCOS contain high levels of aromatase activity in vitro; EGF and/or TGF-α obviously suppressed estradiol production in granulose cells of ovaries from PCOS patients.
