摘要
目的:对皮肤干细胞的生物学特性、分化调控等方面研究成果进行分析。资料来源:应用计算机检索Eldrvier,SpringerLink,KluwerOnlineJour-nals等数据库中1970-01/2005-12相关皮肤干细胞的文献,检索词“skinstemcells;Biologicalcharacteristic;Regulationofdifferentiation”,并限定文献语言种类为English。同时计算机检索中国期刊网2000-01/2005-12相关皮肤干细胞方面的文献,检索词为“皮肤干细胞,生物学特性,分化调控”,并限定语言种类为中文。资料选择:对资料进行初审,选取包括皮肤干细胞的文献,开始查找全文。纳入标准:①皮肤干细胞的来源及生物学特性。②皮肤干细胞的分化调控。排除标准:综述文献、Meta分析、重复研究类文章。资料提炼:共收集到67篇关于皮肤干细胞的文献,纳入21篇符合标准的文献。资料综合:皮肤干细胞是目前研究较多的一种成体干细胞,既有成体干细胞的一般特性,又有其独有的特性。皮肤干细胞具有慢周期性和高度的增殖潜能。皮肤干细胞的生长与分化受转录因子、端粒酶、信号转导通路、整合素、细胞外基质、细胞分泌因子等因素的调控,整合素、角蛋白、p63转录因子等分子标记有助于对皮肤干细胞进行分离、培养和纯化。但皮肤干细胞的发育调控十分复杂,增殖分化条件尚未完全掌握,限制了皮肤类干细胞的研究及临床应用。结论:对皮肤类干细胞进行分离纯化对于皮肤遗传性疾病的基因治疗、组织器官再生、皮肤重建等有重要作用,但在临床应用中仍有许多问题需要解决。
OBJECTIVE: To analyze the study result of biological characteristic and regulation of differentiation of skin stem cells.
DATA SOURCES: We searched Eldrvier,SpringerLink,Kluwer Online Journals for the skin stem cells-related articles published between January 1970 and December 2005 on the computer , with the key words of "skin stem cells; Biological characteristic;Regulation of differentiation" in English. Meanwhile, We also searched the net of Chinese Journal for skin stem cell related articles published between January 2000 and December 2005, with the key words of “skin stem cells; Biological characteristic;Regulation of differentiation” in Chinese.
STUDY SELECTION: The articles were chosen primarily, those were related to skin stem cells were enrolled and their full text were searched for. Inclusive criteria: ①The source and biological characteristic of skin stem cells. ② Molecule regulation of differentiation of skin stem cells. Exclusive criteria: review, Meta analysis and repeated articles were excluded. DATA.EXTRACTION: Totally 67 articles related to skin stem cells were collectgd and 21 of them met the inclusive criteria.
DATA SYNTHESIS: Skin stem cells studied much are adult stem cells with general and unique characteristic. Stem skin stem cells have chronic periodicity and high proliferative potential. The growth and differentiation of skin stem cells were regulated by transcription factor, telomerase, signal tranoduction pathway, integrin, 'extracellular matrix, cellular excreted factor and other factors. The molecular mark of integrin, keratin and p63 transcription factor are helpful to perform isolation, culture and purification of skin stem cells . But the very complex development regulation of skin stem Cells and not mastered proliferative and differentiative conditions limit the study and clinical application of skin stem cells.
CONCLUSION: Isolation and purification of skin stem cells is of very importance in the gene therapy, regeneration of tissue organ, skin reconstruction of skin genetic disease. However, many problems needs to solve in clinical application.
