陆地棉一个丝氨酸/苏氨酸激酶蛋白基因的克隆与表达分析 被引量:13

摘要 通过RTPCR获得一个编码棉花丝氨酸/苏氨酸激酶类似蛋白全长的cDNA片段,其编码的氨基酸序列与拟南芥ATPK3的相似度达到82%,这个基因暂被命名为GhPK1。基因全编码区包括1038个核苷酸,编码346个氨基酸的蛋白。GhPK1的编码产物在265~270个氡基酸处有一个跨膜区并可能结合在内质网膜上。GhPK1在种子和纤维中的表达水平较其它组织高。另外,GhPK1的表达量在受到盐胁迫后上升,说明GhPK1可能参与了盐胁迫反应。 The cDNA encoding the full length of the serine/threonine protein kinase like protein of cotton was cloned through RT-PCR. The amino acid sequence encoded by it shows an 82 % homology to the ATPK3 of Arabidopsis. This gene was tentatively designated as GhPK1. The entire coding region of GhPK1 is 1038 bp and encodes a polypeptide of 346 amino acids. Its product may possess one transmembrane domain in the region of 265 to 270 amino acids and may bind to the membrane of the endoplasmic reticulum. GhPK1 shows a higher expression level in seed and fiber than in other organs. In addition, the expression of GhPK1 is increased by salt induction which indicates GhPK1 may participate in salt-stress reactions.
