摘要
用S-24和网纹香两个哈密瓜品种为材料,研究不同外植体和基本培养基、激素、蔗糖等因子对不定芽诱导率的影响,结果表明:1)两品种不同外植体的不定芽诱导率为子叶柄>子叶>茎尖>真叶;胚轴和根不能诱导不定芽,其中子叶柄是最适外植体,5~8日龄效果较好。2)在6-BA、zT、KT、2ip和2,4-D五种激素中,6-BA的诱导效果最好,其最适浓度为0.5mg/L。3)LS培养基能显著提高两个品种的诱导率和生活力。4)培养基中附加ABA 1.0mg/L可显著提高S-24的诱导率,而附加ABA 2.0mg/L和马铃薯提取液10g/L可显著促进网纹香不定芽的诱导效果。
The sterilized seedlings of two muskmelon varieties ( S-24 and Wang-wenxiang) were used as cultural materials and the effects of different explants, cotyledon peltiole days and culture factors (basal medium, hormone, sucrose, etc. ) on the induction rate of adventitious shoots were studied. The results showed: 1) Among different explants of two varieties, the sequence of induction rate of adventitious shoots was cotyledon peltiole > cotyledon > apex > leaf; the young cotyledon peltiole cultured for 8 days possessed the highest induction rate. 2) Effect of 6-BA on the induction of adventitious shoots was the best in 5 hormones ( 6-BA, ZT, KT, 2, 4-D and 2ip) and the most suitable concentration of 6-BA was 0.5mg/ L. 3) LS medium raised'remarkably the induction rate and vitality of shoots of two varieties. 4) Some additional factors, such as ABA, sucrose and potato juice, could promote the induction of adventitious shoots. Effects of these factors on inducing adventitious shoots were very different with the varieties. Adding ABA 1.0mg/ L in the medium, the induction rate of S-24 was increased. When the medium was supplemented with ABA 2.0mg/ L and potato juice 10g/ L, the induction rate of Wangwenxiang was increased.
