摘要 目的:观察己酮可可碱(PTX)对大鼠胰腺纤维化的影响。方法:大鼠胰管内注射2%三硝基苯磺酸(TNBS),制备大鼠胰腺纤维化模型。将大鼠12只随机分为PTX干预组、对照组。PTX干预组于术后第2天给予PTX 6 mg/(kg.d)腹腔注射,4周后收集两组胰腺标本。采用免疫组化技术检测治疗前后胰腺组织中α-平滑肌肌动蛋白(-αSMA)、血小板衍化生长因子(PDGF)、Ⅰ型胶原(COLⅠ)、磷酸化细胞外信号调节激酶1(P-ERK1)的含量同时做组织胶原纤维染色,部分胰腺组织液氮冻存,以Western blot方法检测-αSMA的表达。结果:对照组发生胰腺纤维化,PTX干预组有轻度纤维化。PTX干预组-αSMA、PDGF、COLⅠ的含量以及P-ERK1在基质细胞中的表达均低于对照组(P<0.01)。胶原纤维染色显示,PTX治疗组胶原面积百分比明显低于对照组(P<0.01)。结论:PTX有抗大鼠胰腺纤维化的作用。 Objective: To observe the effects of pentoxifylline on pancreas fibrosis in rat. Methods: TNBS in a concentration of 2% was injected into the rats'pancreatic duct to set up the model of pancreatitis. Rats of interventional group were treated with FIX, and the expression of PDGF, α-SMA, COL Ⅰ and P-ERK1 were determined by using immunohistochemistry technique and HE and Masson staining in some samples 4 weeks later, meanwhile the expression of α-SMA was detected by using Western blotting. Results: Pancreas fibrosis was observed in control group, whereas it was not significant in FIX treatment group. Compared with the control, FIX significantly reduced both the content of α-SMA, PDGF, COL Ⅰ and P-ERK1 ( P 〈 0.01)and the pancreas fibrosis proportion( P 〈 0.01). Conclusion: Pancreas fibrosis can be inhibited by FIX in rats.
