摘要
目的研究p16在人类非小细胞肺癌中的表达意义及与肿瘤细胞增殖的关系。方法应用免疫组织化学(S-P法)检测了80例非小细胞肺癌组织标本和40例肺良性病变组织中p16和细胞增殖核抗原(PCNA)的表达水平。结果(1)非小细胞肺癌组织中p16表达水平(57.5%)显著低于肺良性病变组织(90.0%)(P<0.01);(2)非小细胞肺癌组织中p16基因表达水平降低与肺癌组织学类型,细胞分化程度,患者P-TNM分期,是否有淋巴结转移存在相关性(P<0.05);(3)非小细胞肺癌组织中p16与PCNA表达呈负相关。结论p16基因表达下调与非小细胞肺癌细胞增殖有关,与肿瘤的发生、发展关系密切。
Objective To investigate the significance of p16 expression and the relationship between the expression of p16 and the proliferation. Method The expressions of p16 and PCNA were detected in 80 nsclc samples and 40 lung benign lesion tissues as control by immunohistochemistry(S-P). Results (1)The expression level of pl 6 gene was found to be significantly lower in NSCLC tissues than in benign lesion tissues(P 〈 0.01 ) ;(2 )The expression level of p16 gene was closely related to histological classification, cancer cell differentiation, P-TNM stages and lymph node involvement in lung cancer patients(P 〈 0.05) ;(3)The expression level of PCNA was found to be significantly higher in NSCLC tissues than in benign lesion tissues( P 〈 0.01 ) ; ( 4 ) The reduction of p16 gene expression might be related with the over-proliferation of lung cancer cell(P 〈 0.01). Conclusion The reduction of p16 gene expression is one of the molecular basis of the over-proliferation in NSCLC cell; p16 gene may be related to the pathogenesis and development of lung cancer.
