用反相高效液相色谱法测定血清中甲硝唑浓度，其操作要点是：样品血清２００μｌ内加入内标工作液及ｐＨ７．５磷酸盐缓冲液后，用３ｍｌ含５％异丙醇的氯仿提取。采用ＹＷＧ－Ｃ１８Ｈ３７分析柱，以甲醇：水（２７：７３）为流动相，茶碱作内标，在２５４ｎｍ波长下监测洗提液，按内标法定量。本法具有快速、简便、灵敏、准确等特点，适合于甲硝唑血药浓度测定及药代动力学研究用。

This paper report a sensitive and rapidmethod for the determination of metronidazole(MTZ)using theophylline as the internal standard. High per-formance liquid chromatograph model 344(Beckman)with a 254nm wavelength UV detector and YWG-C18H37 column (10μm,250 ×4.6mm)was used.Tothe serum sample 200μl,100μl phosphate buffer(0.8mol/L, pH7.5)was added,then extracted with3ml chloroform containing 5%isopropyl alcohol. Theorganic layer was removed and evaporated to drynessunder an air stream in a 40℃ water bath.The residuewas dissolved in 30μl mobile phase and 20μl injected, The mobile phase of water-methanol(73 : 27)waspurnped at 1.0ml/min through the column. The detec-tor operated at 0. 005 aufs,The retention tirnes forMTZ and theophylline were 5.78 and 6. 81 min respec-tively.Standard curve was linear in the concentrationrange of 0. 3125 to 20mg/L. The detection limit inserum was 0.02mg/L. Extraction recovery was 77%-82%; method recovery 99%-102%,withinday RSDless than 3.0%;inter-day RSD less than 3.5%.
