人红细胞血型A抗原的亲和层析纯化

PURIFICATION OF HUMAN RED BLOOD CELL GROUP-A PROTEIN BY AFFINITY CHROMATOGRAPHY

ＡＢＯ抗原是人红细胞上一类重要的血型抗原，抗Ａ、Ｂ型抗体在临床检验中是必不 可少的试剂．为了提高抗Ａ单克隆抗体的生产效率，用ＩｇＭ抗体新和层析法提纯了Ａ糖脂蛋白的 可溶性部分，并在用一些免疫生化的方法加以鉴定之后，将它通过抗体分子的中介与胶颗粒连结， 成为颗粒性抗原免疫Ｂａｌｂ／ｃ小鼠，以探讨这种纯化和免疫方法的可行性。

The blood group antigens ABO are important human blood-type antigens,and their antibodies are commonly used in clinical pharmacology. To enhance the efficiency of the production of anti-A monoclonal antibodies,purified the soluble human blood group-A protein with IgM affinity chromatography. After identified it with several immunological biochemical methods,conjugated it with antibody molecule mediated by affinity gel granule,then,immunized Balb/c mouse with this kind of granule antigen. Finally,discussed the meanings of this method of immunization.
