摘要
目的探讨高压氧(HBO)暴露对淋巴细胞功能的影响。方法将.Balb/c小鼠随机分成正常对照组、HBO组、肿瘤组、肿瘤HBO组。用接种S-180腹水癌细胞的方法制成免疫功能低下动物模型。经HBO暴露,取血标本观察淋巴细胞-肿瘤细胞花环率、淋巴细胞数、淋巴细胞百分比;用ELISA双抗夹心法测定小鼠血清IgG浓度;用3H-TdR掺入法测定脾淋巴细胞增殖;用同位素靶细胞释放法观察 NK细胞杀伤活性和LAK细胞活性;取脾脏观察小鼠脾指数的变化。结果 (1)淋巴细胞-肿瘤细胞花环率肿瘤HBO组明显高于对照组(P<0.01)。(2)淋巴细胞计数各组之间均无明显变化。(3)淋巴细胞百分比对照组分别比肿瘤HBO组和肿瘤组高(P<0.01,P<0.05),HBO组比肿瘤HBO组高(P< O.01)。(4)IgG测定各组间无统计学差异。(5)淋巴细胞增殖HBO组比对照组低(P<0.01),肿瘤 HBO组比HBO组低(P<0.01),肿瘤组比HBO组低(P<0.01)。(6)NK细胞活性肿瘤组比对照组低 (P<0.05),肿瘤HBO组比肿瘤组高(P<0.05),HBO组比对照组有降低的趋势(P=0.07)。(7)LAK 细胞活性肿瘤组比对照组高(P<0.05)。(8)脾指数各组之间差异无统计学意义(P>0.05)。结论压力为0.2 MPa,氧体积分数为87%的HBO暴露和/或肿瘤接种对免疫系统的作用是多方面的,有的部分增强,有的部分减弱,值得进一步研究。
Objective To explore the influence of hyperbaric oxygenation (HBO) on the lymphocyte functions of tumor-bearing mice. Methods Tumor-bearing mice models were established by injecting S-180 cells to male Balb/c mice. After HBO exposure, blood samples were taken and lymphocyte-tumor cell rosette rates, lymphocyte count and lymphocyte percentage were observed. The IgG concentration in the blood serum was determined by ELISA method; the proliferation of spleen lymphocytes was measured by ^3H-TDR uptake test; the cytotoxicity of nature killer cells and LAK cells was observed by target cell release method and the spleen index was detected. Results The lymphocyte-tumor cell rosette rate in the HBO-tumor group was significantly higher than that of the control group ( P 〈 0.01 ) ; no significant difference in lymphocyte counts was shown in different groups ; the lymphocyte percentage in the control group was higher than that in the HBO-tumor group and the tumor group ( P 〈 0.01, P 〈 0.05 ), and that in the HBO group was higher than that in the HBO-tumor group (P 〈 0.01 ); no statistical significance in the IgG concentration was observed in different groups; the proliferation of lymphocytes in HBO group and in tumor group was lower than that in the control group (P 〈 0.01 ), and that in the HBO-tumor group was lower than that in tumor group (P 〈 0.01 ) and in HBO group (P 〈0.01 ) ; the cytotoxicity of nature killer cells in the control group was higher than that in the tumor group ( P 〈 0.05 ) ; the cytotoxicity of LAK cells in tumor group was higher than that in the control group (P 〈 0. 05 ) ; no statistical significance in spleen index was observed in different groups ( P 〈 0. 05 ). Conclusions HBO (87% 02,0.2MPa) had various effects on immune system, strengthening some functions and suppressing some functions.
